SiliaBond Carbonate amide synthesis workup with microwave

The use of functionalized silica allows to greatly simplify not only organic synthesis but also purification. In some cases, it permits to reduce the work up to a simple filtration and evaporation of the solvent. With the advent of microwave synthesizers, reaction times have been tremendously reduced from hours and days to only minutes. These facts made it obvious that both technologies, SiliaBond silicas and microwave synthesizers, could be combined to prepare pure products in small scale, and this, in only minutes.

As we are developing more products and applications for amide synthesis, it became clear that SiliaBond Carbonate could play a key role in further simplifying the work up in amide coupling reactions. This is the reason why we worked on and presented a poster studying this subject at the 229th National ACS Meeting in San Diego.

This poster presented different examples and results on the use of SiliaBond Carbonate in the synthesis of amides using other SiliaBond reagents. We investigated its efficiency as a scavenger of various coupling reagents namely 1-hydroxybenzotriazole (HOBt), pentafluorophenol, 4-nitrophenol, 1-hydroxy-7-azabenzotriazole (HOAt), and N-hydroxysuccinimide (HOSu) and compared it with 3 suppliers of polymer-supported carbonate. We also looked at the yields and purities of different amide syntheses using SiliaBond Carbodiimide and Carbonate. In these reactions, the only work up consisted in a filtration and evaporation of the solvent. Lastly, we looked at the ability of SiliaBond Carbonate to neutralize amine salts of various compositions. Here is a glimpse of the results obtained:

FLEXI-TIER : 96-Position Collection Plates and Accessories

Our Flexi-Tier™ 96-position plate has a unique design formaximum flexibility. It can have a fixed height or can be“flexi-tiered” to various heights to accommodate differentvial sizes. The plate can be built using spacers to hold 0.5,1.0, 1.5 and 2mL capacities (depending on the height limi-tations of the instrument) and can hold various length glassor polypropylene 9mm diameter conical or flat bottomvials. The Flexi-Tier™ system fits in all autosamplers orrobotic systems.

Flexi-Tier™ plates have an open bottom enabling it to beimmersed in heating or cooling liquids allowing total fluidcontact with the vials. This permits uniform transfer of heator cold to and from the vials (the plate does notfloat in thewater bath).

F l e x i - T i e r ™ plates have a standard 8x12 position format with an alpha-numeric I.D. grid. The plate cover locks the liner in place preventing vial pullout and provides a secondary sealing system. The secure fit of the vials in the base plate permits use with shakers and vortex mixers during sample preparation.
